The seed
Karoo first appears in Ch. 109 (page 4) with caption-tag KAROO / THE ULTRA SPOTBILL. His introduction is comedic-incompetent — he sits on Mr. 9 by mistake, flies the wrong direction during Peacock Slasher, and gets yelled at by Vivi for poor target-acquisition.
The payoff (pending)
Subsequent arcs (Little Garden, Drum, Alabasta) reveal Karoo as earnest combat-tier mount — outruns camels across the Alabasta desert, carries crucial intelligence between Alabasta cities, and remains in Vivi’s service permanently after the Straw Hats depart. Karoo also serves as the emotional-companion mascot of the Alabasta arc.
Why this matters
Karoo’s comedic-incompetent-debut-then-earnest-loyalty-reveal is a classic Oda animal-companion template (cf. Chouchou the dog in Orange Town, the Lapahn rabbits in Drum, the Camel in Alabasta). The debut-as-buffoon-then-reveal-as-loyal pattern is structural foreshadowing of the same template Vivi herself undergoes — Ms. Wednesday is introduced as a comedic-villain officer-disguise, then revealed as a loyal-royal-character. Karoo and Vivi share a first-impression-inverts-into-true-character register.
