The seed
Ch. 45 page 8, Gin’s warning to Luffy: “The seas are vast. The Grand Line is just one narrow route. There are plenty of safer waters to plunder. You’re young. Don’t rush to your doom.” The Grand-Line-as-narrow-road geography is articulated by an East Blue pirate-officer for the first time.
Why this matters
The one narrow route phrasing is geographically literal — the Grand Line is one specific sea-lane, not a region. The Ch. 100-102 Reverse Mountain entry will pay off this exact geography. Pre-Ch. 100, the Grand Line is repeatedly described as a region by less-informed characters; Gin’s route framing is the first correct description on-page.
Cross-reference: Grand Line events ledger — the Ch. 22 Nami-draws-the-world panel (pp. 14-15) is the closest prior approximation, but Nami’s framing is map-as-diagram rather than the in-character narrow-route phrasing Gin uses. Gin’s phrasing is the first direct-speech articulation of the route geography by an in-world character.
